Background

DNM1L is a key protein involved in mitochondrial and peroxisomal division. It mediates membrane fission by oligomerizing into membrane-associated tubular structures that constrict and sever mitochondrial membranes through a GTP hydrolysis-dependent mechanism (PubMed:23530241, PubMed:23584531, PubMed:33850055). Recruitment to scission sites is facilitated by membrane receptors such as MFF, MIEF1, and MIEF2 (PubMed:23283981, PubMed:23921378, PubMed:29899447). GTP hydrolysis triggers dissociation from receptors, allowing DNM1L filaments to form closed rings likely sufficient for membrane scission (PubMed:29899447). It acts downstream of PINK1 to promote mitochondrial fission in a PRKN-dependent manner (PubMed:32484300) and regulates mitochondrial division during mitosis (PubMed:19411255, PubMed:26992161, PubMed:27301544, PubMed:27328748). By suppressing oxidative damage, it ensures the survival of certain postmitotic neurons, including Purkinje cells (By similarity). Essential for normal brain development, particularly in the cerebellum (PubMed:17460227, PubMed:26992161, PubMed:27145208, PubMed:27301544, PubMed:27328748), it also participates in developmentally regulated apoptosis during neural tube formation (By similarity). Rhythmic phosphorylation at Ser-637 is critical for circadian control of mitochondrial ATP production (PubMed:29478834). Overexpression inhibits peroxisomal division.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
