PDK1 Antibody (YA236) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to PDK1.

PDK1, a pivotal kinase, exerts significant control over glucose and fatty acid metabolism by phosphorylating the pyruvate dehydrogenase subunits PDHA1 and PDHA2. This phosphorylation event acts as a molecular switch, inhibiting pyruvate dehydrogenase activity and finely tuning metabolite flux through the tricarboxylic acid cycle. Consequently, PDK1 down-regulates aerobic respiration and impedes the conversion of pyruvate to acetyl-coenzyme A. Beyond its metabolic functions, PDK1 emerges as a critical player in cellular responses to hypoxia, where it plays a key role in promoting cell proliferation under oxygen-deprived conditions.   Western blot analysis of extracts from Hela (lane 1) and HepG2 (lane 2) using PDK1 antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
