Background

PARK7 is a multifunctional protein with controversial molecular functions, primarily acting as an oxidative stress sensor, redox-sensitive chaperone, and protease to protect cells from oxidative stress and cell death. It is involved in neuroprotective mechanisms, such as stabilizing NFE2L2 and PINK1 proteins, positively regulating the androgen signaling pathway for male fertility, and influencing cell growth and transformation through pathways like NF-kappa-B signaling modulation. Described as a protein and nucleotide deglycase, PARK7 catalyzes the deglycation of Maillard adducts formed between amino groups of proteins/nucleotides and reactive carbonyl groups of glyoxals, though this role is debated. As a protein deglycase, it repairs methylglyoxal- and glyoxal-glycated proteins, releasing repaired proteins along with lactate or glycolate, and reactivates proteins by reversing glycation on cysteine, arginine, and lysine residues. It also functions as a nucleotide deglycase, repairing glycated guanine in free nucleotides (GTP, GDP, GMP, dGTP) and nucleic acids, participating in the guanine glycation repair (GG repair) system. PARK7 protects histones from methylglyoxal adduction, modulates chromatin arginine modifications, and restores histone 3 function by removing glycations. It exhibits minimal glyoxalase activity, potentially linked to deglycase function, and eliminates hydrogen peroxide to prevent cell death. Essential for mitochondrial morphology, function, and autophagy, PARK7 regulates mitochondrial ROS levels and glucose homeostasis in pancreatic islets (by similar). It binds mRNAs with GG/CC motifs to suppress translation (reversibly under oxidative stress) and acts as a metal-binding protein, countering copper/mercury toxicity. In macrophages, it interacts with NCF1 to direct NADPH oxidase-dependent ROS production, offering sepsis protection (by similar).
