Background

MAPK14 is a serine/threonine kinase that serves as a core component of the MAP kinase signaling pathway. As one of the four p38 MAPKs, it plays a pivotal role in cellular response cascades triggered by extracellular stimuli like pro-inflammatory cytokines or physical stress, directly activating transcription factors. p38 MAPKs phosphorylate approximately 200-300 substrates, including downstream kinases (e.g., RPS6KA5/MSK1 and RPS6KA4/MSK2, which phosphorylate transcription factors such as CREB1, ATF1, RELA/NFKB3, STAT1, STAT3, as well as histone H3 and nucleosomal protein HMGN1). Other kinase targets like MAPKAPK2/MK2 and MAPKAPK3/MK3 regulate post-transcriptional gene expression by phosphorylating ZFP36 and ELAVL1. by similar: MAPK14 also activates MKNK1/MNK1 and MKNK2/MNK2 to phosphorylate translation initiation factor EIF4E2, and interacts with casein kinase II to activate TP53/p53. In the cytoplasm, it regulates protein degradation via phosphorylating CFLAR and SIAH2, while inhibiting autophagy by disrupting ATG9 trafficking. Additionally, MAPK14 controls receptor endocytosis by phosphorylating EGFR and RAB5A effectors, and activates ADAM17-mediated TGF-α shedding to stimulate EGFR signaling. In the nucleus, it phosphorylates transcription factors (e.g., ATF2, ELK1, TP53) and enhances chromatin accessibility of inflammatory genes (e.g., IL6, IL8) via histone H3 Ser-10 phosphorylation (H3S10ph). Kinase-independent roles include modulating O-GlcNAcylation through OGT interaction. During development, MAPK14 regulates EPO expression for placental angiogenesis and stress erythropoiesis. Isoform MXI2 is activated by oxidative stress, while EXIP may initiate apoptosis. During microbial infection, M.tuberculosis EsxA activates MAPK14 in T-cells to suppress IFN-γ production.
