P2RY8 is a G protein-coupled receptor that specifically binds S-geranylgeranyl-glutathione (GGG), an endogenous metabolite present in lymphoid tissues. It couples GGG binding to GNA13 activation and downstream suppression of AKT signaling, thereby regulating lymphocyte positioning and growth within lymphoid organs. In lymphoid follicles, P2RY8 responds to local GGG gradients established by GGT5 (via GGG catabolism) and ABCC1 (via extracellular transport), confining B cells and follicular helper T cells within germinal centers (GCs), where lower GGG concentrations are found around the follicular dendritic cell network (by similar). In the bone marrow, it similarly restricts chemotactic transmigration of B cells, T cells, and NK cells from blood vessels to the parenchyma via GGT5- and ABCC1-mediated GGG gradients (by similar). Additionally, it contributes to a GNA13-dependent pathway that suppresses GC B cell growth.
