CDKN2A/p14ARF is a tumor suppressor capable of inducing cell cycle arrest in G1 and G2 phases. It binds to MDM2 and sequesters it in the nucleolus, blocking its nucleocytoplasmic shuttling, thereby inhibiting MDM2-induced p53 degradation and enhancing p53-dependent transactivation and apoptosis. Additionally, CDKN2A/p14ARF induces G2 arrest and apoptosis in a p53-independent manner by preventing cyclin B1/CDC2 complex activation. It binds to BCL6, E2F1, and MYC, blocking their transcriptional activator activity (but not affecting MYC's transcriptional repression). CDKN2A/p14ARF interacts with TOP1/TOPOI, stimulating its activity; this complex binds to rRNA gene promoters and may play a role in rRNA transcription and/or maturation. It also interacts with NPM1/B23, promoting its polyubiquitination and degradation, thereby inhibiting rRNA processing. Furthermore, it may inhibit ribosome biogenesis by binding to the nucleolar localization sequence of TTF1, preventing its nucleolar localization (By similarity). CDKN2A/p14ARF interacts with COMMD1, promoting its 'Lys63'-linked polyubiquitination; binds to UBE2I/UBC9, enhancing sumoylation of partners like MDM2 and E2F1; and binds to HUWE1, repressing its ubiquitin ligase activity. It may regulate cell proliferation and apoptosis during mammary gland development and be involved in autophagy and caspase-independent cell death regulation. The short-lived mitochondrial isoform is stabilized by C1QBP.
