Background

OXGR1 is a G protein-coupled receptor that primarily recognizes dicarboxylates and amino dicarboxylates. It serves as a receptor for itaconate, a metabolite produced by myeloid lineages. In the respiratory epithelium, OXGR1 couples itaconate binding to the activation of GNA11 and subsequent intracellular Ca2+ release, facilitating mucociliary clearance of airborne pathogens. Additionally, OXGR1 acts as a high-affinity receptor for leukotriene E4 (LTE4), which is produced by mast cells during allergic inflammation, triggering mucin release from pulmonary epithelium in response to fungal allergens and regulating goblet cell homeostasis (By similarity). In the kidney, OXGR1 functions as a receptor for alpha-ketoglutarate, produced by proximal tubule renal cells during metabolic alkalosis. It binds alpha-ketoglutarate to drive transepithelial salt reabsorption and bicarbonate secretion by SLC26A4/pendrin-positive intercalated cells, participating in an intrarenal paracrine signaling pathway (By similarity).
