Osteocalcin Antibody is an unconjugated, approximately 11 kDa, rabbit-derived, anti-Osteocalcin polyclonal antibody. Osteocalcin Antibody can be used for: WB, ELISA, IHC-P, IHC-F, IF expriments in human, mouse, and predicted: rat background without labeling.

Osteocalcin is a bone protein constituting 1-2% of total bone protein and acts as a negative regulator of bone formation. It limits bone formation without impairing bone resorption or mineralization (By similarity) and binds strongly to apatite and calcium. The uncarboxylated form functions as a hormone secreted by osteoblasts, regulating various cellular processes such as energy metabolism, male fertility, and brain development. It modulates energy metabolism by promoting pancreatic beta-cell proliferation, insulin secretion and sensitivity, and energy expenditure. Uncarboxylated osteocalcin also enhances testosterone production in the testes by acting as a ligand for the G protein-coupled receptor GPRC6A on Leydig cells, initiating a signaling cascade that upregulates enzymes for testosterone synthesis in a CREB-dependent manner. Additionally, osteocalcin regulates brain development by crossing the blood-brain barrier and binding to GPR158 on neurons, triggering a signaling response that prevents hippocampal neuronal apoptosis, promotes monoamine neurotransmitter synthesis, and inhibits GABA production. Maternal osteocalcin crosses the placenta and is essential for fetal brain development.

  Western blot analysis of extracts from M-muscle (lane 1) and K562 (lane 2) and M-lung (lane 3) and M-lymph (lane 4) using Osteocalcin antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
