Background

NUMA1 is a microtubule (MT)-binding protein that plays a crucial role in the formation and maintenance of spindle poles and the alignment and segregation of chromosomes during mitotic cell division. It tethers the minus ends of MTs at spindle poles, which is essential for spindle pole establishment and stability. NUMA1 regulates mitotic spindle orientation during metaphase and elongation during anaphase in a dynein-dynactin-dependent manner. During metaphase, it forms a ternary complex with GPSM2 and G(i) alpha proteins to recruit and anchor the dynein-dynactin complex at the cortical regions above spindle poles, ensuring proper spindle orientation. In anaphase, NUMA1 mediates dynein-dynactin accumulation at the polar cortex by binding phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2), contributing to spindle elongation and chromosome segregation. It also interacts with other phosphoinositides, including PIP, LPA, and PIP3. NUMA1 is involved in asymmetric cell division, anastral spindle assembly, and positively regulates TNKS localization to spindle poles. As a major nuclear matrix component, it may serve a structural role in non-mitotic contexts. Additionally, NUMA1 is required for epidermal differentiation and hair follicle morphogenesis (by similar).
