Nucleophosmin is a multifunctional protein involved in diverse cellular processes including ribosome biogenesis, centrosome duplication, protein chaperoning, histone assembly, cell proliferation, and regulation of tumor suppressors p53/TP53 and ARF. It binds ribosomes to facilitate ribosome nuclear export and associates with nucleolar ribonucleoprotein structures while interacting with single-stranded nucleic acids. Acting as a chaperonin, it assists in the assembly of core histones H3, H2B, and H4. Nucleophosmin stimulates APEX1 endonuclease activity on apurinic/apyrimidinic (AP) double-stranded DNA but inhibits its activity on AP single-stranded RNA, potentially regulating APEX1-mediated repair of AP sites in rDNA and removal of oxidized rRNA within nucleoli. In concert with BRCA2, it regulates centrosome duplication, and PLK2 phosphorylation triggers centriole replication. It negatively regulates EIF2AK2/PKR activation by suppressing its autophosphorylation to inhibit apoptosis, counteracts ATF5-mediated cell proliferation suppression, and relieves ATF5-induced G2/M blockade. When complexed with MYC, it enhances transcription of MYC target genes. By similarity, it may function as a chaperonin or cotransporter in the nucleolar localization of transcription termination factor TTF1.
