NQO1 Antibody (YA697) is a Mouse-derived and non-conjugated IgG2b monoclonal antibody, targeting to NQO1.

NQO1 is a flavin-containing quinone reductase that catalyzes the two-electron reduction of quinones to hydroquinones using NADH or NADPH as electron donors. It operates via a ping-pong kinetic mechanism, sequentially transferring electrons from NAD(P)H to the flavin cofactor and then to the quinone substrate, thereby bypassing the formation of semiquinone intermediates and reactive oxygen species (By similarity). Its primary role involves regulating cellular redox homeostasis through quinone detoxification, reducing components of the plasma membrane redox system (e.g., coenzyme Q and vitamin quinones) to generate antioxidant hydroquinone forms, while potentially scavenging superoxide to prevent hydroquinone oxidation and facilitate excretion. Paradoxically, it can also activate certain quinones by generating redox-reactive hydroquinones with DNA cross-linking antitumor properties. Additionally, NQO1 functions as a gatekeeper of the 20S proteasome core; under oxidative stress, it interacts with tumor suppressors TP53 and TP73 in an NADH-dependent manner to inhibit their ubiquitin-independent degradation by the 20S proteasome.

  Western blot analysis of extracts from A549 (lane 2(20μg), SiHa (lane 3(20μg), Hela (lane 4(20μg) using NQO1 Antibody (HY-P80250) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80250, 1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
