Nicastrin Antibody (YA1397) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Nicastrin.

The related gene encodes a type I transmembrane glycoprotein that is an integral component of the multimeric gamma-secretase complex. The encoded protein cleaves integral membrane proteins, including Notch receptors and beta-amyloid precursor protein, and may be a stabilizing cofactor required for gamma-secretase complex assembly. The cleavage of beta-amyloid precursor protein yields amyloid beta peptide, the main component of the neuritic plaque and the hallmark lesion in the brains of patients with Alzheimer's disease; however, the nature of the encoded protein's role in Alzheimer's disease is not known for certain. Mutations in The related gene are associated with familial acne inversa. A pseudorelated gene of The related gene is present on chromosome 21. Alternatively spliced transcript variants of The related gene have been described, but the full-length nature of some of these variants has not been determined.

  Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), K562 (lane 2, 15 μg), U-87MG (lane 3, 15 μg), and C6 (lane 4, 15 μg) using Nicastrin Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80487, 1:40000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
