NCoA-3 is a nuclear receptor coactivator that directly binds nuclear receptors and stimulates transcriptional activity in a hormone-dependent manner. It plays a central role in forming a multisubunit coactivator complex, likely acting through chromatin remodeling. NCoA-3 is involved in coactivating various nuclear receptors, including those for steroids (GR and ER), retinoids (RARs and RXRs), thyroid hormone (TRs), vitamin D3 (VDR), and prostanoids (PPARs). It exhibits histone acetyltransferase activity and also participates in coactivating the NF-kappa-B pathway via interaction with the NFKB1 subunit.
