mtTFA is a mitochondrial transcription regulatory protein that binds to the mitochondrial light strand promoter and functions in mitochondrial transcription regulation. It is a core component of the mitochondrial transcription initiation complex, which minimally includes TFB2M, TFAM, and POLRMT, and is essential for basal transcription of mitochondrial DNA (mtDNA). Within this complex, TFAM recruits POLRMT to specific promoter regions, while TFB2M induces structural changes in POLRMT to facilitate promoter opening and non-template strand trapping. mtTFA promotes transcription initiation from the HSP1 and light strand promoters by binding upstream of transcriptional start sites and exhibits DNA unwinding activity. Its HMG boxes bend the mitochondrial light strand promoter DNA into a U-turn conformation.
