Description

MRE11 Antibody (YA283) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to MRE11.

Background

Mre11 is the core component of the MRN complex, playing a central role in double-strand break (DSB) repair, DNA recombination, telomere integrity maintenance, and meiosis. The MRN complex repairs DSBs via homologous recombination (HR), primarily during the S and G2 phases, by (1) mediating end resection of damaged DNA to generate single-stranded DNA (ssDNA), a key initial step in HR, and (2) recruiting other repair factors and activating ATM and ATR in response to DNA damage. MRE11 possesses both single-strand endonuclease and double-strand-specific 3'-5' exonuclease activities, collaborating with RBBP8/CtIP at DSB sites to initiate end resection. It first cleaves the 5' strand endonucleolytically to suppress non-homologous end joining (NHEJ) and license HR, then generates an ssDNA gap via 3' to 5' exonucleolytic degradation to enable EXO1- and DNA2-mediated long-range resection. RBBP8/CtIP enhances MRE11's endonuclease activity to clear protein-DNA adducts (By similar). The MRN complex also activates ATM and ATR for DNA damage signaling and processes R-loops. In telomeres, it may modulate t-loop formation. MRE11 contains two DNA-binding domains (DBDs) for binding both ssDNA and dsDNA.
