MEK-4 is a transcription factor essential for the commitment of lymphoid progenitors to the B-lymphocyte lineage. It functions dually by repressing B-lineage inappropriate genes while activating B-lineage-specific genes. This regulation impacts cell adhesion and migration, induces V(H)-to-D(H)J(H) recombination, facilitates pre-B-cell receptor signaling, and promotes development to the mature B-cell stage. Additionally, MEK-4 represses the cohesin-release factor WAPL, leading to global chromosomal architecture changes in pro-B cells that enable diverse antibody repertoire generation. In microbial infection contexts, MEK-4 maintains Epstein-Barr virus genome copy number by supporting EBNA1/oriP-dependent binding and transcription, while also inhibiting lytic EBV reactivation through modulation of viral BZLF1 activity.
