MCM3 is a component of the MCM2-7 complex (MCM complex), the replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. As a core component of the CDC45-MCM-GINS (CMG) helicase, it forms part of the molecular machine that unwinds template DNA during replication and serves as the scaffold for replisome assembly. The active ATPase sites in the MCM2-7 ring are generated through interfaces of adjacent subunits, with a conserved arginine finger motif provided in trans relative to the Walker A box ATP-binding site of the neighboring subunit. The six ATPase active sites likely contribute differentially to the helicase activity. MCM3 is required for S phase entry and cell division (Probable).
