Background

MCM2 is a core component of the MCM2-7 complex (MCM complex), the replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. It forms part of the CDC45-MCM-GINS (CMG) helicase, the molecular machine that unwinds template DNA during replication and serves as the scaffold for replisome assembly (by similar: PubMed:32453425, PubMed:34694004, PubMed:34700328, PubMed:35585232). The ATPase active sites in the MCM2-7 ring are generated through interfaces of adjacent subunits, with a conserved arginine finger motif acting in trans on the Walker A box ATP-binding site of neighboring subunits, though the six ATPase sites likely contribute asymmetrically to helicase activity (by similar: PubMed:32453425). MCM2 is required for S-phase entry and cell division (by similar: PubMed:8175912) and also regulates terminally differentiated cochlear hair cell development and apoptosis induction (by similar: PubMed:26196677).
