MBNL1 Antibody (YA2601) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to MBNL1.

Background

MBNL1 is an RNA-binding protein that mediates pre-mRNA alternative splicing regulation, functioning as either an activator or repressor for specific pre-mRNA targets. In muscle tissue, it inhibits cardiac troponin-T (TNNT2) pre-mRNA exon inclusion while promoting insulin receptor (IR) pre-mRNA exon inclusion. It regulates TNNT2 exon 5 skipping by competing with U2AF2 and binds to the stem-loop structure within the polypyrimidine tract of TNNT2 intron 4 during spliceosome assembly, thereby inhibiting the formation of the spliceosomal A complex on TNNT2 pre-mRNA intron 4. MBNL1 recognizes the 5'-YGCU(U/G)Y-3' consensus sequence and binds to IR RNA as well as expanded CUG repeat RNA (which forms a hairpin structure with GC base pairs and bulged, unpaired U residues). It cooperates with RNA-binding proteins RBPMS and RBFOX2 to activate alternative splicing events in vascular smooth muscle cells. Additionally, MBNL1 regulates NCOR2 alternative splicing (By similarity).
