Background

LIM Kinase 1 (LIMK1) is a serine/threonine-protein kinase that plays a pivotal role in regulating actin filament dynamics. It acts downstream of multiple Rho family GTPase signaling pathways and is activated by upstream kinases such as ROCK1, PAK1, and PAK4, which phosphorylate a threonine residue in its activation loop. Activated LIMK1 phosphorylates and inactivates actin-binding/depolymerizing factors cofilin-1 (CFL1), cofilin-2 (CFL2), and destrin (DSTN), thereby inhibiting the severing of filamentous actin (F-actin) and stabilizing the actin cytoskeleton. Through this mechanism, LIMK1 regulates various actin-dependent biological processes, including cell motility, cell cycle progression, and differentiation. Additionally, LIMK1 phosphorylates TPPP on serine residues, promoting microtubule disassembly, and may contribute to brain development by stimulating axonal outgrowth. By similar: LIMK1 exhibits a dominant-negative effect on actin cytoskeletal changes and is required for atypical chemokine receptor ACKR2-induced phosphorylation of cofilin (CFL1).
