Ku-80 Antibody (YA4943) is a mouse-derived and non-conjugated monoclonal antibody, targeting to Ku-80. It can be applicated for WB, IHC-P, ICC/IF, FC, ELISA assays, in the background of human, mouse.

Ku80 is a single-stranded DNA-dependent ATP-dependent helicase that plays a key role in DNA non-homologous end joining (NHEJ) by recruiting DNA-PK to DNA. It is required for double-strand break repair and V(D)J recombination, and also has a role in chromosome translocation. The DNA helicase II complex binds preferentially to fork-like ends of double-stranded DNA in a cell cycle-dependent manner and works in the 3'-5' direction. During NHEJ, the XRCC5-XRRC6 dimer performs the recognition step by binding to broken DNA ends and protecting them from further resection. This dimer acts as a regulatory subunit of the DNA-PK complex, increasing the affinity of the catalytic subunit PRKDC to DNA. It likely stabilizes broken DNA ends and brings them together. The XRCC5-XRRC6 dimer may also function as a 5'-deoxyribose-5-phosphate lyase (5'-dRP lyase), catalyzing the beta-elimination of 5' deoxyribose-5-phosphate at abasic sites near double-strand breaks. XRCC5 probably serves as the catalytic subunit for 5'-dRP activity, cleaning termini of abasic sites before broken ends can be joined. Together with APEX1, the dimer acts as a negative transcriptional regulator, while in association with NAA15, it activates osteocalcin expression. As part of the DNA-PK complex, Ku80 is involved in ribosome assembly by promoting rRNA processing. It also plays a role in DNA virus-mediated innate immune response by assembling into the HDP-RNP complex, which serves as a platform for IRF3 phosphorylation and activation of the cGAS-STING pathway.
