KSHV ORF45 Antibody (YA4607)

KSHV ORF45 Antibody (YA4607) is a mouse-derived and non-conjugated IgG1 antibody, targeting to KSHV ORF45. It can be applicated for ICC, ELISA assays, in the background of virus.

KSHV ORF45 prevents the establishment of cellular antiviral state by blocking virus-induced phosphorylation and activation of host interferon regulatory factor 7 (IRF7), a transcription factor critical for the induction of interferons alpha and beta. Mechanistically, ORF45 acts as an alternative substrate, competing with IRF7 and inhibiting its phosphorylation by IKBKE or TBK1. Additionally, ORF45 activates the NLRP1 inflammasome by interacting with the N-terminal part of host NLRP1, promoting its translocation into the nucleus, thereby relieving autoinhibition and leading to inflammasome activation. It also promotes late transcription and translation of viral lytic genes by constitutively activating host extracellular signal-regulated kinase (ERK)-p90 ribosomal S6 kinase (RPS6KA1). Furthermore, ORF45 supports viral replication by modulating the host p53 (TP53) signaling pathway, interacting with p53 and preventing its binding to the deubiquitinase USP7, leading to cytoplasmic sequestration of p53 and reduced transcriptional activity.
