Background

KRIT1 is a component of the CCM signaling pathway, which plays a pivotal role in regulating heart and vessel formation and integrity (By similarity). As a negative regulator of angiogenesis, it inhibits endothelial cell proliferation, apoptosis, migration, lumen formation, and sprouting angiogenesis. KRIT1 promotes AKT phosphorylation via both NOTCH-dependent and independent mechanisms while indirectly suppressing ERK1/2 phosphorylation through activation of the DELTA-NOTCH cascade. In collaboration with CDH5, it establishes and maintains endothelial cell polarity and vascular lumen by recruiting and activating the Par polarity complex and RAP1B. KRIT1 is essential for localizing phosphorylated PRKCZ, PARD3, TIAM1, and RAP1B to cell junctions, ensuring junctional stability. It modulates integrin signaling by binding ITGB1BP1, thereby disrupting ITGB1-ITGB1BP1 interaction. As a microtubule-associated protein, KRIT1 binds PIP2-containing membranes in a GTP-bound RAP1-dependent manner. It maintains intracellular reactive oxygen species (ROS) homeostasis via an antioxidant pathway involving FOXO1 and SOD2, facilitating cyclin-D1 (CCND1) downregulation for cell cycle quiescence. Additionally, KRIT1 may regulate macroautophagy by suppressing the mTOR pathway.
