KHDRBS2 is an RNA-binding protein involved in regulating alternative splicing, influencing mRNA splice site selection and exon inclusion. It binds to both poly(A) and poly(U) homopolymers (by similarity). By directly binding to purine-rich exonic enhancers, KHDRBS2 promotes concentration-dependent exon inclusion in CD44 pre-mRNA. This protein also regulates alternative splicing of NRXN1 in the laminin G-like domain 6 containing the evolutionarily conserved neurexin alternative spliced segment 4 (AS4), which participates in neurexin's selective targeting to postsynaptic partners. KHDRBS2 mediates cell-type specific alternative splicing of NRXN1 at AS4 and acts synergistically with SAM68 to promote exon skipping, while exhibiting antagonistic effects with SAM68 in NRXN3 exon skipping at AS4 (by similarity). Its splice site selection regulatory ability is inhibited by FYN-mediated phosphorylation. Additionally, it may function as an adapter protein for Src kinases during mitosis.
