JAM3 is a junctional adhesion protein that mediates heterotypic cell-cell interactions with its cognate receptor JAM2 to regulate various cellular processes. It plays a role in the homing and mobilization of hematopoietic stem and progenitor cells within the bone marrow, contributing to their retention via JAM3 expressed on bone marrow stromal cells. JAM3 is central to leukocyte extravasation by facilitating transmigration through the endothelium (by similar). During spermatogenesis, JAM2 and JAM3, expressed by Sertoli and germ cells respectively, mediate interactions between these cell types, essential for germ cell anchorage to Sertoli cells and the assembly of cell polarity complexes during spermatid differentiation (by similar). Additionally, JAM3 functions as a counter-receptor for ITGAM, mediating leukocyte-platelet interactions and regulating transepithelial migration of polymorphonuclear neutrophils (PMN). It is involved in angiogenesis and the regulation of cell migration (Probable). In myogenesis, JAM3 participates in myocyte fusion (by similar) and promotes chemotaxis of vascular endothelial cells, stimulating angiogenesis.
