Background

ITCH is an E3 ubiquitin-protein ligase that accepts ubiquitin from an E2 ubiquitin-conjugating enzyme as a thioester and directly transfers it to target substrates. It catalyzes 'Lys-29'-, 'Lys-48'-, and 'Lys-63'-linked ubiquitin conjugation and regulates inflammatory signaling pathways. As a core component of a ubiquitin-editing complex with TNFAIP3, TAX1BP1, and RNF11, it ensures transient inflammatory signaling by promoting TNFAIP3-mediated deubiquitination of 'Lys-63' chains on RIPK1 and subsequent 'Lys-48'-linked polyubiquitination, leading to RIPK1 proteasomal degradation and termination of NFKB1 activation. ITCH also ubiquitinates RIPK2 via 'Lys-63' linkages, modulating NOD2-dependent signaling, and influences transcription factor activity, likely playing a key role in immune response regulation. Additionally, ITCH ubiquitinates NFE2 via 'Lys-63' linkages, impacting hematopoietic lineage development. Similar functions include mediating JUN and JUNB ubiquitination and degradation (by similar) and negatively regulating MAVS-dependent antiviral responses by 'Lys-48'-linked ubiquitination and degradation of MAVS. ITCH regulates Wnt receptor FZD4 sorting to degradative endocytic pathways by modulating PI4K2A activity and ubiquitinates CXCR4 and ESCRT-0 components HGS and STAM. It also targets DTX1 for lysosomal degradation and controls NOTCH1 degradation via 'Lys-29'-linked polyubiquitination. ITCH ubiquitinates SNX9 and MAP3K7 (by similar) and, with UBR5, regulates apoptosis and reactive oxygen species levels through branched ubiquitination of TXNIP (ITCH synthesizes 'Lys-63' chains, while UBR5 branches 'Lys-48' chains). ITCH mediates epidermal growth factor's antiapoptotic effects by ubiquitinating and degrading p15 BID and enhances BRAT1 ubiquitination in the presence of NDFIP1. Furthermore, ITCH inhibits influenza A virus replication by 'Lys-48'-linked ubiquitination and proteasomal degradation of viral M1 protein. Lastly, ITCH ubiquitinates NEDD9/HEF1, leading to its proteasomal degradation.
