IRAK-4 Antibody (YA4923) is a mouse-derived and non-conjugated monoclonal antibody, targeting to IRAK-4. It can be applicated for WB, IHC-P, ICC/IF, FC, ELISA assays, in the background of human, mouse, monkey.

Background

IRAK-4 is a serine/threonine-protein kinase that plays a critical role in initiating innate immune responses against foreign pathogens. It is involved in Toll-like receptor (TLR) and IL-1R signaling pathways and is rapidly recruited by MYD88 to the receptor-signaling complex upon TLR activation, forming the Myddosome together with IRAK2. IRAK-4 phosphorylates IRAK1, stimulating its kinase activity and intensive autophosphorylation. It also phosphorylates E3 ubiquitin ligases Pellino proteins (PELI1, PELI2, and PELI3) to promote pellino-mediated polyubiquitination of IRAK1. The ubiquitin-binding domain of IKBKG/NEMO then binds to polyubiquitinated IRAK1, bringing together the IRAK1-MAP3K7/TAK1-TRAF6 complex and the NEMO-IKKA-IKKB complex. Subsequently, MAP3K7/TAK1 activates IKKs (CHUK/IKKA and IKBKB/IKKB), leading to NF-kappa-B nuclear translocation and activation. Additionally, IRAK-4 phosphorylates TIRAP to promote its ubiquitination and subsequent degradation. By similar mechanisms (by similar), IRAK-4 phosphorylates NCF1 and regulates NADPH oxidase activation after LPS stimulation, suggesting a comparable role during microbial infections.
