IL17A is an effector cytokine of the innate and adaptive immune systems involved in antimicrobial host defense and maintenance of tissue integrity. It signals through the IL17RA-IL17RC heterodimeric receptor complex, triggering homotypic interaction between IL17RA and IL17RC chains with the TRAF3IP2 adapter, leading to TRAF6-mediated activation of NF-kappa-B and MAP kinase pathways. This results in transcriptional activation of cytokines, chemokines, antimicrobial peptides, and matrix metalloproteinases, potentially inducing strong immune inflammation. As a signature effector cytokine of T-helper 17 (Th17) cells, IL17A primarily induces neutrophil activation and recruitment at infection and inflammatory sites. In airway epithelium, it mediates neutrophil chemotaxis via CXCL1 and CXCL5 induction. In secondary lymphoid organs, it contributes to germinal center formation by regulating B cell chemotactic responses to CXCL12 and CXCL13, enhancing B cell retention, somatic hypermutation, and plasma cell differentiation. IL17A also acts as an effector cytokine in gamma-delta T cells, functioning downstream of IL1B, TLR2, and IL23A-IL12B to promote neutrophil recruitment for bacterial clearance. It plays a role in epithelial barrier integrity, regulating OCLN localization and tight junction biogenesis during acute injury. In synergy with IL17F, it induces mucosal epithelial cells to produce antimicrobial beta-defensins (DEFB1, DEFB103A, DEFB104A), limiting microbial invasion. Additionally, IL17A contributes to antiviral defense by enhancing T cell cytotoxicity against West Nile virus, promoting B cell recruitment in H5N1 influenza infection, and driving B-1a B cell differentiation for H1N1 clearance via virus-specific IgM production.
