Background

IL-17 Receptor A (IL-17RA) is a receptor for IL17A and IL17F, key effector cytokines of the innate and adaptive immune systems involved in antimicrobial host defense and tissue integrity maintenance. It binds IL17A with higher affinity than IL17F and forms a heterodimeric complex with IL17RC to interact with IL17A or IL17F homodimers or IL17A/IL17F heterodimers. Cytokine binding triggers homotypic interaction between IL17RA and IL17RC chains with the TRAF3IP2 adapter, leading to TRAF6-mediated activation of NF-kappa-B and MAP kinase pathways, ultimately inducing transcriptional activation of cytokines, chemokines, antimicrobial peptides, and matrix metalloproteinases, potentially driving robust immune inflammation. In antimicrobial defense, IL-17RA primarily promotes neutrophil activation and recruitment to infection sites to eliminate extracellular bacteria and fungi (by similar). In secondary lymphoid organs, it contributes to germinal center formation by modulating B cell chemotaxis toward CXCL12 and CXCL13, enhancing B cell retention in germinal centers, somatic hypermutation rates, and plasma cell differentiation (by similar). It also maintains epithelial barrier integrity during homeostasis and infection by stimulating mucosal epithelial cells to produce antimicrobial β-defensins (DEFB1, DEFB103A, and DEFB104A), limiting microbial invasion (by similar). In antiviral defense, IL-17RA enhances immunity through mechanisms such as promoting T cell cytotoxicity against West Nile virus or driving B-1a B cell differentiation to generate virus-specific IgM antibodies for influenza clearance (by similar). IL-17RA additionally serves as a receptor for IL17C in a heterodimeric complex with IL17RE. Furthermore, SARS-CoV-2 ORF8 protein activates the IL-17 pathway via IL-17RA, increasing pro-inflammatory factor secretion through NF-kappa-B signaling.
