IGFBP2 is a multifunctional protein that critically regulates the availability of IGFs (e.g., IGF1 and IGF2) to their receptors, thereby modulating IGF-mediated cellular processes such as proliferation, differentiation, and apoptosis in a cell-type-specific manner. It coordinates with receptor protein tyrosine phosphatase beta/PTPRB and the IGF1 receptor to regulate IGF1-mediated signaling by stimulating PTEN phosphorylation, leading to its inactivation and AKT1 activation. Additionally, IGFBP2 promotes cell migration via interaction with integrin alpha5/ITGA5 through an RGD motif, while its binding to ITGA5/ITGB1 enhances endothelial progenitor cell adhesion to endothelial cells.
