Background

IDE (Insulin-Degrading Enzyme) is an enzyme involved in the cellular degradation of various peptides, including insulin, APP peptides, IAPP peptides, natriuretic peptides, glucagon, bradykinin, kallidin, and others, thereby playing a role in intercellular peptide signaling. Substrate binding induces significant conformational changes, enabling the degradation of larger substrates like insulin. Additionally, IDE contributes to the regulation of peptide hormone signaling cascades and blood glucose homeostasis by degrading insulin, glucagon, and IAPP (by similar). It also participates in the degradation and clearance of APP-derived amyloidogenic peptides secreted by neurons and microglia. IDE degrades natriuretic peptides (ANP, BNP, and CNP), inactivating their ability to elevate intracellular cGMP, and degrades an aberrant frameshifted form of NPPA (fsNPPA) associated with familial atrial fibrillation. Furthermore, IDE is involved in antigen processing, generating both the N- and C-termini of the MAGEA3-derived antigenic peptide (EVDPIGHLY) presented to cytotoxic T lymphocytes via MHC class I. In microbial infections, the membrane-associated isoform of IDE acts as an entry receptor for varicella-zoster virus (VZV).
