Background

HSP70 is a molecular chaperone implicated in diverse cellular processes, including proteome stress protection, folding/transport of nascent polypeptides, degradation of misfolded proteins, and modulation of protein complex dynamics. It plays a central role in protein quality control by orchestrating ATP-dependent cycles of substrate binding/release, facilitated by co-chaperones that regulate its ATPase activity (e.g., HSP40), nucleotide exchange (e.g., BAG1/2/3), or substrate fate (e.g., HOPX/STUB1). Its substrate affinity is nucleotide-dependent: low in ATP-bound state and high upon ATP hydrolysis to ADP. Acetylation switches its function between refolding (via HOPX) and degradation (via STUB1). HSP70 maintains mitotic centrosome integrity, promotes STUB1-mediated ubiquitination of SMAD3, FOXP3 (in Tregs), and NFATC3 (by similarity), suppresses HSF1 activity post-heat shock, and aids clearance of misfolded PRDM1. During rotavirus infection, it serves as a post-attachment entry receptor.
