Hsp70 is a crucial cytoprotective protein upregulated in response to temperature increases and various stresses. Its gene expression is mediated by the transcription factor HSF1, which binds to HSE elements in the hsp70 promoter under stress. As the most ancient anti-stress defense system, HSP70 promotes tumor cell survival by interacting with multiple points in apoptotic signaling pathways. It functions as a cellular lifeguard with anti-apoptotic effects, modulates intrinsic and extrinsic apoptotic signaling, and regulates caspase-independent apoptosis in cancer cells. Due to its anti-cancer and immunogenic properties, HSP70 inhibitors are being sought, though preclinical and clinical evaluations lag behind Hsp90 inhibitors currently in phase II/III trials. Extracellular HSP70 is immunogenic, acting as an adjuvant that, combined with chaperones, may aid vaccine production and cancer treatment. Elevated extracellular HSP70 indicates poor cancer prognosis. In syngeneic mice, HSP70 inhibition or depletion induces tumor regression, anti-tumor immune responses, and target cell apoptosis. HSP70 is an essential chaperone class that maintains proteostasis by interacting with client proteins, playing key roles in stress survival and thermotolerance development. Its function relies on a two-domain structure comprising an N-terminal nucleotide-binding domain (NBD) and a substrate-binding domain (SBD), with substrate affinity regulated by ATP hydrolysis and ADP release cycles, acting as a nucleotide-regulated clamp for transiently binding misfolded proteins.
