HSC70 Antibody (YA5396) is a mouse-derived and non-conjugated monoclonal antibody, targeting to HSC70. It can be applicated for IHC-P, WB assays, in the background of human, mouse, rat.

Background

HSC70 is a molecular chaperone involved in diverse cellular processes, including proteome stress protection, folding/transport of newly synthesized polypeptides, chaperone-mediated autophagy, degradation of misfolded proteins, assembly/disassembly of protein complexes, and antigen presentation. It plays a central role in protein quality control by ensuring proper folding, refolding of misfolded proteins, and targeting damaged proteins for degradation. This function is driven by ATP binding, hydrolysis, and ADP release cycles regulated by co-chaperones. HSC70's substrate affinity is nucleotide-dependent: low in ATP-bound state and high after ATP-to-ADP hydrolysis. Its co-chaperones fall into three classes: J-domain HSP40s (stimulating ATPase activity), nucleotide exchange factors (e.g., BAG1/2/3 promoting ADP-to-ATP transition), and TPR-domain chaperones (e.g., HOPX/STUB1). HSC70 also mediates mitochondrial import via TOMM70, represses transcriptional activation, participates in spliceosome assembly as part of the PRP19-CDC5L complex, binds LPS to trigger inflammatory responses, recognizes KFERQ-motif proteins for lysosomal degradation in CMA, facilitates MHC class II antigen presentation with LAMP2, contributes to ERAD pathway with STUB1, and promotes clathrin uncoating of vesicles via DNAJC6 interaction (by similarity).
