hnRNP U Antibody (YA5127) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to hnRNP U.

Background

hnRNP U is a DNA- and RNA-binding protein involved in various cellular processes, including nuclear chromatin organization, telomere-length regulation, transcription, mRNA alternative splicing and stability, Xist-mediated transcriptional silencing, and mitotic cell progression. It regulates interphase large-scale gene-rich chromatin organization through chromatin-associated RNAs (caRNAs) in a transcription-dependent manner, maintaining genomic stability. hnRNP U is required for the localization of the long non-coding Xist RNA on the inactive X chromosome (Xi) and the initiation and maintenance of X-linked transcriptional gene silencing during X-inactivation (By similarity). It acts as an RNA polymerase II (Pol II) holoenzyme transcription regulator, promoting transcription initiation by associating with the core-TFIIH basal transcription factor complex in an actin-dependent manner (PubMed). hnRNP U inhibits Pol II transcription elongation by blocking C-terminal domain (CTD) phosphorylation and dissociates from the Pol II pre-initiation complex before productive elongation (PubMed). It positively regulates CBX5-induced transcriptional silencing and nuclear retention of CBX5 (PubMed) and negatively regulates glucocorticoid-mediated transcriptional activation (PubMed). In embryonic stem cells, hnRNP U is a key regulator of transcription initiation and elongation under leukemia inhibitory factor (LIF) signaling (By similarity). It participates in long non-coding RNA H19-mediated Pol II transcriptional repression (PubMed) and the circadian regulation of BMAL1 transcription (By similarity). hnRNP U modulates global pre-mRNA alternative splicing by regulating U2 small nuclear ribonucleoprotein (snRNP) biogenesis (PubMed) and contributes to mRNA stability (PubMed). During mitotic cell cycle regulation, it facilitates stable kinetochore-microtubule (MT) attachment, spindle organization, and chromosome congression (PubMed). hnRNP U binds to double- and single-stranded DNA and RNA, as well as various oligonucleotides (PubMed), and interacts with chromatin-associated RNAs (caRNAs) and Xist RNA (PubMed). In microbial infection, hnRNP U negatively regulates HIV-1 replication by preventing cytoplasmic accumulation of viral mRNA transcripts.
