HLA E Antibody (YA3437) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to HLA E.

Background

HLA E is a non-classical major histocompatibility class Ib molecule involved in immune self-nonself discrimination. It forms a complex with B2M/beta-2-microglobulin to bind nonamer self-peptides derived from the signal sequence of classical MHC class Ia molecules (VL9 peptides - VMAPRT[V/L][L/V/I/F]L). The peptide-bound HLA-E-B2M heterotrimeric complex primarily functions as a ligand for the natural killer (NK) cell inhibitory receptor KLRD1-KLRC1, enabling NK cells to monitor the expression of other MHC class I molecules in healthy cells and tolerate self. Under cellular stress, HLA E preferentially binds signal sequence-derived peptides from stress-induced chaperones and is no longer recognized by the NK cell inhibitory receptor KLRD1-KLRC1, impairing protection from NK cells. Additionally, HLA E binds signal sequence-derived peptides from non-classical MHC class Ib HLA-G molecules and acts as a ligand for the NK cell activating receptor KLRD1-KLRC2, likely playing roles in adaptive NK cell generation, effector functions, and maternal-fetal tolerance during pregnancy. Beyond self-peptides, HLA E can also bind and present pathogen-derived peptides conformationally similar to VL9 peptides to unconventional CD8-positive cytotoxic T cells via alpha-beta T cell receptor (TCR), triggering antimicrobial immune responses. It presents HIV gag peptides (immunodominant KAFSPEVIPMF and subdominant KALGPAATL epitopes) to CD8-positive T cell clones expressing TRAV17-containing TCR, inducing HLA-E-restricted T cell responses. Furthermore, HLA E presents mycobacterial peptides to HLA-E-restricted CD8-positive T cells, eliciting cytotoxic and immunoregulatory functions. In viral infections, pathogens like human cytomegalovirus exploit HLA E by downregulating host MHC class I molecules and binding viral peptides to HLA E, restoring its expression and inducing HLA-E-dependent NK cell tolerance to infected cells. HLA E may also bind HIV-1 gag/Capsid protein p24-derived peptide (AISPRTLNA) to inhibit NK cell cytotoxicity, aiding viral immune evasion. During SARS-CoV-2 infection, HLA E binds the SARS-CoV-2 S/Spike protein S1-derived peptide (LQPRTFLL), contributing to NK cell exhaustion and dampened antiviral immune surveillance.
