HIPK2 acts as a negative regulator through the phosphorylation-induced proteasomal degradation of CTNNB1 and the antiapoptotic factor CTBP1. Additionally, it serves as an intermediate kinase in the Wnt/beta-catenin signaling pathway, promoting the proteasomal degradation of MYB. HIPK2's intricate involvement in cellular responses includes transcriptional co-suppression of HIF1A in response to hypoxia and the activation of TP73. Furthermore, it plays a crucial role in DNA damage responses, stabilizing PML, triggering ZBTB4 protein degradation, and preventing DAZAP2-dependent ubiquitination of HIPK2. Moreover, HIPK2 contributes to angiogenesis, erythroid differentiation, and the regulation of eye development during late embryogenesis.

  Western blot analysis was performed on extracts from HepG2 (lane 1, 15 μg), and Mouse heart (lane 2, 15 μg) using HIPK2 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
