GTF2H1 is a component of the general transcription and DNA repair factor IIH (TFIIH) core complex, involved in both general and transcription-coupled nucleotide excision repair (NER) of damaged DNA and, when complexed with CAK, in RNA transcription by RNA polymerase II. In NER, TFIIH opens the DNA around the lesion to facilitate the excision of the damaged oligonucleotide and its replacement with a new DNA fragment. In transcription, TFIIH plays a crucial role in transcription initiation. After the pre-initiation complex (PIC) is established, TFIIH is required for promoter opening and promoter escape. The phosphorylation of the C-terminal tail (CTD) of the largest subunit of RNA polymerase II by the CAK kinase module regulates transcription initiation.
