Background

GABA A Receptor alpha 1 is the alpha subunit of the heteropentameric ligand-gated chloride channel activated by Gamma-aminobutyric acid (GABA), a major inhibitory neurotransmitter in the brain. GABA-gated chloride channels, also known as GABA(A) receptors (GABAAR), consist of five subunits arranged around a central pore, with GABA active binding sites located at the interface of alpha and beta subunits. Upon GABA activation, GABAARs selectively permit chloride ion flow across the cell membrane along their electrochemical gradient (By similarity). Alpha-1/GABRA1-containing GABAARs are predominantly synaptic (By similarity). Chloride influx into postsynaptic neurons following GABAAR activation reduces their ability to generate new action potentials, thereby inhibiting nerve transmission (By similarity). GABAARs containing alpha-1 and beta-2 or beta-3 subunits exhibit synaptogenic activity, with the gamma-2 subunit being necessary but not sufficient for rapid synaptic contact formation. GABAARs also function as histamine receptors, where histamine binds at the interface of two neighboring beta subunits, potentiating GABA responses (By similarity). GABAARs composed of alpha, beta, and epsilon subunits allow spontaneous chloride channel activity while maintaining structural integrity for GABA-gated openings (By similarity). Alpha-1-mediated plasticity in the orbitofrontal cortex regulates context-dependent action selection. Together with rho subunits, it may also modulate neuronal and glial GABAergic transmission in the cerebellum.
