FXR1 is an mRNA-binding protein that regulates mRNA translation and/or stability, playing essential roles in neurogenesis, muscle development, and spermatogenesis. It specifically binds AU-rich elements (AREs) in the 3'-UTR of target mRNAs and undergoes liquid-liquid phase separation to assemble mRNAs into cytoplasmic ribonucleoprotein granules (By similar). During late spermatogenesis, FXR1 activates translation of stored mRNAs by recruiting the translation initiation factor EIF4G3 (By similar). It promotes MYC transcript translation via eIF4F complex recruitment and suppresses inflammation by destabilizing pro-inflammatory mRNAs (e.g., TNF mRNA) (By similar). In neurons, FXR1 inhibits GRIA2/GluA2 translation to modulate synaptic plasticity and enhances CDKN1A mRNA expression to regulate neural stem cell proliferation (By similar). It also contributes to muscle tissue development and prevents cytoplasmic aggregation of nucleoporins to maintain nuclear pore complex localization. FXR1 stabilizes PKP2 mRNA through interaction with PKP3 and potentially influences PKP1, PKP3, and DSP via similar mechanisms. Additionally, it forms a cytoplasmic messenger ribonucleoprotein (mRNP) network by packaging long mRNAs, serving as a scaffold to facilitate signaling processes such as actomyosin reorganization.
