Background

FOXA2 is a transcription factor involved in embryonic development, establishment of tissue-specific gene expression, and regulation of gene expression in differentiated tissues. It acts as a 'pioneer' factor by opening compacted chromatin for other proteins through interactions with nucleosomal core histones, thereby replacing linker histones at target enhancer and/or promoter sites. FOXA2 binds DNA with the consensus sequence 5'-[AC]A[AT]T[AG]TT[GT][AG][CT]T[CT]-3' (By similarity). During embryonic development, it is required for notochord formation and plays a role in the development of multiple endoderm-derived organ systems, including the liver, pancreas, and lungs, with FOXA1 and FOXA2 exhibiting partially redundant functions. Initially identified as a transcriptional activator for liver-specific genes such as AFP, albumin, tyrosine aminotransferase, and PEPCK, FOXA2 interacts with their cis-regulatory regions. It also contributes to glucose homeostasis by regulating genes critical for glucose sensing in pancreatic beta-cells and glucose metabolism. Additionally, FOXA2 is involved in fat metabolism regulation and binds to the fibrinogen beta promoter, participating in IL6-induced transcriptional activation of fibrinogen beta.
