Fibrillarin is an S-adenosyl-L-methionine-dependent methyltransferase capable of methylating both RNAs and proteins. It participates in pre-rRNA processing by catalyzing site-specific 2'-hydroxyl methylation of ribose moieties in pre-ribosomal RNA (By similar: site specificity is determined by a guide RNA that base-pairs with the substrate, with methylation occurring at a characteristic distance from the guide RNA pairing sequence). It likely mediates 2'-O-methylation of U6 snRNAs in box C/D RNP complexes, a modification essential for mRNA splicing fidelity. Additionally, it functions as a protein methyltransferase by methylating Gln-105 of histone H2A (H2AQ104me), a modification that impairs FACT complex binding and is specifically localized at the 35S ribosomal DNA locus. As a component of the small subunit (SSU) processome (the first precursor of the eukaryotic small ribosomal subunit), it collaborates with numerous ribosome biogenesis factors, RNA chaperones, and ribosomal proteins during nucleolar SSU processome assembly to coordinate RNA folding, modifications, rearrangements, cleavage, and targeted pre-ribosomal RNA degradation via the RNA exosome.
