FGFR3 Antibody (YA438) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to FGFR3.

FGFR3 is a tyrosine-protein kinase that functions as a cell-surface receptor for fibroblast growth factors (FGFs) and plays essential roles in regulating cell proliferation, differentiation, and apoptosis. It is critically involved in chondrocyte differentiation, proliferation, and apoptosis, and is required for normal skeletal development. The protein regulates both osteogenesis and postnatal bone mineralization by osteoblasts, exhibiting dual functions of promoting chondrocyte apoptosis while potentially enhancing cancer cell proliferation. FGFR3 is necessary for inner ear development and phosphorylates PLCG1, CBL, and FRS2. Ligand binding activates multiple signaling pathways: PLCG1 activation generates diacylglycerol and inositol 1,4,5-trisphosphate, while FRS2 phosphorylation mediates recruitment of GRB2, GAB1, PIK3R1, and SOS1 to activate RAS, MAPK1/ERK2, MAPK3/ERK1, MAP kinase pathways, and the AKT1 signaling pathway. It also participates in vitamin D metabolism regulation. Mutations causing constitutive kinase activation or impairing FGFR3 maturation, internalization, and degradation lead to aberrant signaling. Overexpressed or constitutively active FGFR3 promotes activation of PTPN11/SHP2, STAT1, STAT5A, and STAT5B. The secreted isoform 3 retains FGF1/FGF2 binding capacity and may interfere with FGF signaling.

  Western blot analysis of extracts from HepG2 (lane 1) and M-lymph (lane 2) and K562 (lane 3) using FGFR3 antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
