FAK (Focal Adhesion Kinase) is a non-receptor protein-tyrosine kinase that plays pivotal roles in regulating cell migration, adhesion, spreading, actin cytoskeleton reorganization, formation/disassembly of focal adhesions and protrusions, cell cycle progression, proliferation, and apoptosis. It is essential for embryonic development, placental formation, angiogenesis, cardiomyocyte migration/proliferation, heart development, axon growth, neuronal migration, synaptogenesis, and nervous system development. FAK also participates in osteoblast differentiation and osteogenesis. It mediates integrin signaling and transduces signals downstream of growth factor receptors, GPCRs, EPHA2, netrin receptors, and LDL receptors. By forming complexes with SRC family kinases, FAK creates phosphorylation-dependent binding sites to regulate PI3K-AKT1, MAPK/ERK cascades, and Rho GTPase activity through GEFs/GAPs. It phosphorylates targets like NEDD9, SRC, ACTN1, and recruits MDM2 to modulate p53 stability. The kinase-deficient FRNK isoform acts as a dominant-negative regulator by suppressing FAK phosphorylation and nuclear LPXN/SRF-dependent transcription.
