GSPT1/ERF3A delivers ETF1/ERF1 to stop codons: The eRF1-eRF3-GTP complex binds to a stop codon in the ribosomal A-site, and GTP hydrolysis by GSPT1/ERF3A induces a conformational change leading to its dissociation, allowing ETF1/ERF1 to fully occupy the A-site. Additionally, eRF3a is part of the transient SURF complex that recruits UPF1 to stalled ribosomes during nonsense-mediated decay (NMD) of mRNAs containing premature stop codons.   Western blot analysis was performed on extracts from HepG2 (lane 1, 15 μg), Hela (lane 2, 15 μg), 3T3 (lane 3, 15 μg), and 293 (lane 4, 15 μg) using eRF3a Rabbit pAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80487, 1:60000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
