Description

ITK Antibody (YA4827) is a mouse-derived and non-conjugated monoclonal antibody, targeting to Emt. It can be applicated for WB, ICC/IF, FC, ELISA assays, in the background of human.

Background

Emt is a tyrosine kinase essential for regulating adaptive immune responses. It controls the development, function, and differentiation of conventional T-cells and nonconventional NKT-cells. Upon T-cell receptor (TCR) activation by antigen-presenting cells (APC), phosphorylation cascades recruit ITK to the membrane near stimulated TCR, where LCK phosphorylates it, triggering autophosphorylation and full activation. Activated ITK phosphorylates PLCG1, activating this lipase to cleave its substrates, leading to calcium release from the endoplasmic reticulum and nuclear translocation of NFAT for transcriptional activity. ITK also phosphorylates key adapters LAT and LCP2, recruiting signaling molecules like VAV1 to drive lymphokine production, T-cell proliferation, and differentiation. It is required for TCR-mediated calcium responses in γδ T-cells and may modulate transcriptomic signatures in immature Vγ2-positive γδ T-cell subsets (By similarity). Additionally, ITK phosphorylates TBX21 at Tyr-530, mediating its interaction with GATA3 (By similarity).
