EHMT2 is a histone methyltransferase that specifically mono- and dimethylates 'Lys-9' of histone H3 (H3K9me1 and H3K9me2) in euchromatin. H3K9me serves as a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. Additionally, EHMT2 mediates monomethylation of 'Lys-56' of histone H3 (H3K56me1) during G1 phase, promoting interaction between histone H3 and PCNA and regulating DNA replication. It also weakly methylates 'Lys-27' of histone H3 (H3K27me). EHMT2 is required for DNA methylation, though its histone methyltransferase activity is not necessary for this process, indicating independent functions. It is likely targeted to histone H3 by DNA-binding proteins such as E2F6, MGA, MAX, and/or DP1 and may methylate histone H1. Beyond histone methylation, EHMT2 methylates non-histone proteins, including dimethylation of 'Lys-373' of p53/TP53, and methylates CDYL, WIZ, ACIN1, DNMT1, HDAC1, ERCC6, KLF12, and itself.

  Western blot analysis was performed on extracts from Hela (lane 1, 20 μg), 293(lane 2, 20 μg), and A431 (lane 3, 20 μg) using EHMT2/G9A Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Mouse/Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
