EGR1 Antibody (YA467) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to EGR1.

EGR1 is a transcriptional regulator that recognizes and binds to the DNA sequence 5'-GCG(T/G)GGGCG-3' (EGR-site) in the promoter region of target genes (By similarity). It binds double-stranded target DNA regardless of cytosine methylation status. EGR1 regulates the transcription of numerous target genes, playing a key role in cellular responses to growth factors, DNA damage, and ischemia, as well as in controlling cell survival, proliferation, and death. It activates the expression of p53/TP53 and TGFB1, contributing to tumor prevention. EGR1 is essential for normal mitosis progression and hepatocyte proliferation after partial hepatectomy. It mediates responses to ischemia and hypoxia by regulating the expression of proteins like IL1B and CXCL2, which are involved in inflammation and post-ischemic tissue damage (By similarity). Additionally, EGR1 modulates the amplitude of clock gene expression rhythms (BMAL1, PER2, NR1D1) in the liver via PER1 activation and regulates the rhythmic expression of core-clock gene BMAL1 in the suprachiasmatic nucleus (SCN) (By similarity).

  Western blot analysis of extracts from 293T (lane 1) and THP-1 (lane 2) and Jurkat (lane 3) using EGR1 antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
