Background

DYNLL1 functions as a non-catalytic accessory component of the cytoplasmic dynein 1 complex, facilitating dynein's linkage to cargoes and regulatory adapter proteins (By similarity). This complex acts as a motor for retrograde intracellular transport of vesicles and organelles along microtubules (By similarity) and may modulate cytoskeletal spatial organization (By similarity). Beyond transport roles, DYNLL1 serves as a protein-protein adapter that inhibits/sequesters target proteins. During DNA damage response, phosphorylation at Ser-88 enables TP53BP1-mediated recruitment to double-strand breaks (DSBs), where it disrupts MRE11 dimerization to block DNA end resection; unphosphorylated DYNLL1 promotes Shieldin complex recruitment. It binds and inhibits neuronal nitric oxide synthase/NOS1 activity (By similarity), enhances ESR1 transactivation and nuclear localization, and regulates apoptosis by tethering BCL2L11 to microtubules—upon apoptotic stimuli, the BCL2L11-DYNLL1 complex relocalizes to mitochondria to counteract BCL2's anti-apoptotic function.
