Dynamin II is a GTPase that catalyzes the hydrolysis of GTP and utilizes this energy to mediate vesicle scission at the plasma membrane during endocytosis and filament remodeling at various actin structures during actin cytoskeleton organization. It plays a crucial role in vesicular trafficking processes, including clathrin-mediated endocytosis (CME), exocytic and clathrin-coated vesicle formation from the trans-Golgi network, and PDGF-stimulated macropinocytosis. During vesicular trafficking, Dynamin II associates with the membrane via lipid binding and self-assembles into ring-like structures through oligomerization, forming a helical polymer around the vesicle membrane, ultimately leading to vesicle scission. In actin cytoskeleton organization, it mediates the arrangement of stress fibers and actin bundles in podocytes (by similar). Dynamin II also remodels bundled actin filaments in a GTPase-dependent manner through its interaction with CTTN and plays a role in orchestrating the global actomyosin cytoskeleton (by similar). Additionally, it participates in autophagy by contributing to ATG9A vesicle formation through interaction with SNX18 and mediating recycling endosome scission leading to autophagosome release via MAP1LC3B interaction. It further regulates the maturation of apoptotic cell corpse-containing phagosomes by recruiting PIK3C3 to the phagosome membrane (by similar). Dynamin II is also involved in cytokinesis (by similar), centrosome cohesion (by similar), neuron morphology regulation, axon growth, and neuronal growth cone formation (by similar), as well as membrane tubulation.
